Abstract

Bacteria encounter different kinds of environmental stresses during their life span. Cold-shock stress is one of those. To cope up with the cold-shock stress, bacteria produce cold-shock proteins. CsdA is one of the major cold-shock proteins, which act as RNA helicase to rescue the cell to adapt to the cold-shock condition. But how this gene operates and regulates its expression has not been studied yet. In this thesis, I have described my research work about the identification and characterization of the csdA promoter and its 5'UTR regions in Escherichia coli. In chapter 1, I have described the following findings; We have a promoter less plasmid vector. By using this plasmid vector, I have performed a series of in vitro experiments and identified the csdA promoter of E. coli. Further I have performed in vivo deletion experiments and confirmed the in vitro findings of the promoter region of this csdA gene in E. coli. Furthermore, I have characterized the csdA promoter region. Based on various in vitro and in vivo genetic and biochemical studies described within chapter 1, I have reported about the identification of the true promoter region of the csdA gene of E. coli that it is located more than 800 bp ahead of the translation start site of csdA gene. In this connection, I have revealed the occurrence of an extremely large and possibly very rare 800nt long 5'UTR in bacteria. In the last chapter, i.e., chapter: 2, I have described about my findings of some unique characteristics of bacterial translation factors from extensive data search and analysis.
