Abstract
Leishmaniasis, caused by Leishmania spp. is a neglected tropical disease with severe public health and socioeconomic impacts. The parasite survives and replicates within the acid phagolysosome of host macrophages, where it faces diverse kinds of stresses, including acidic stress. Our lab has provided a putative model of intracellular pH regulation which includes two carbonic anhydrases and a putative bicarbonate transporter. We have identified a probable candidate protein and, in this project, we are trying to decipher the role of this protein in the parasite physiology by generating a knockout of the protein using a recently-developed system of performing CRISPR-Cas9-mediated genome editing in Leishmania.
