Abstract

Enzymatic reactions often drive through charge transition states to exhibit structure function relationship, which stems from higher substrate binding affinity and greater accessibility of noncovalent interactions. In this context, herein, we demonstrate the capabilities of short amyloid peptide assemblies to template water-mediated reduction of ester molecules. A series of seven residue amyloidogenic peptides were designed that upon self-assembly access nanotubular morphologies, accessing para crystalline surfaces with solvent exposed residues. Remarkably, the amyloid nanotubes featuring diverse cationic residues were found to promote ester reduction in the presence of the cofactor. Further, the non-covalent binding abilities of amyloid were utilized to show a controlled reduction reaction which could not be achieved by using standard ester reduction methods. Moreover, the amyloid micro phases thus foreshadow the critical role of electrostatic effect and the generation of binding pockets of extant proteins that may have tremendous potential to contribute towards the development of advanced functional materials.
