Abstract

Psychrotolerant microorganisms have a decent capability to survive in cold environment and face detrimental conditions by using diverse mechanisms which can be applied in agriculture and industries for efficient and environmental friendly production. A multi-step investigation was done to examine the bacteria isolated from high altitude pristine Himalaya water samples. Four of the isolates were undertaken for Physiological and molecular experiments. The isolates were rod-shaped, and two of them were Gram-positive without any oxidase activity detectable, while the other two were Gram-negative with sufficient oxidase activity. The isolates were mostly found to be non-motile and appeared to possess the catalase enzyme. All but one isolate demonstrated the ability to hydrolyse starch. None of the isolates were detected for the tryptophanase enzyme. One of the isolates was found to have the capacity to utilize gelatin. Molecular examination using 16S rDNA PCR found about 1500 bp bands, which were subsequently analysed using Sanger's sequencing method, identifying them to be of the genus Pseudomonas, Ochrobactrum and Brucella, and Rhodococcus. The enzymatic characteristics of these isolates, as well as their capacity to endure stress at low temperatures, may then be further explored in future.
