Abstract

Reversible addition-fragmentation chain transfer (RAFT) approach has been shown to be one of the most versatile methods for producing polymers with well-defined architectures. The RAFT method provides unrivalled flexibility in the synthesis of suitable polymeric architectures for various therapeutic applications. In the recent past, gas therapy has gained much attention for biomedical applications and nitric oxide (NO) is one of the potential candidates. NO acts as a signalling molecule with an extensive range of functions in both health and disease. The deficiency of NO caused in the human body has been an issue for quite a long time. Exogenous NO sources have been proven a very efficient way to supplement NO when the body cannot generate enough NO to work properly. Hence, polymers prepared with RAFT polymerization can provide a desirable platform for NO delivery. NO is one of the smallest pharmacological molecules, with promising anti-restenosis, wound healing, anti-cancer, and anti-bacterial properties. For biological applications, the development of NO-releasing donors became necessary. Rather than concentrating on tiny molecules, we synthesized a photoactive water-soluble polymeric NO donor that releases NO under exposure to light. The goal of this chapter is to highlight not only the fundamental principles and obstacles of NO delivery but also to provide insight into the future development of innovative NO delivery systems for clinical applications.
