Abstract

The conventional means of controlling the recurring pandemics of Type A Influenza Virus (IAV) infections remain unsuccessful primarily because of vaccine failure and surging cases of drug resistance. As an alternative means for developing effective measures to control IAV infection, the exogenous application of cytokines has gained significant attention in recent times. Among them, Type III Interferons, particularly IFNλ3, have been claimed to induce potent anti-viral host responses by upregulating the expression of several immunoregulatory factors that are directly involved in inhibiting virus replication. To this end, we examined the effect of IFNλ3 application on lung tissue in an experimental murine model. Here, we used a modified recombinant L. lactis vector secreting murine Interferon Lambda-3 (MuIFNλ3). To see the direct role of IFNλ3, we performed the transcriptional analysis of several other cytokines and Interferon Stimulatory Genes (ISGs) commonly linked to the IFNλ3 signaling cascade. Our result suggests that recombinant IFNλ3 is secreted by rL. lactis is functional and can enhance key immunoregulatory genes required for inhibiting IAV replication and ensuring effective protection. However, future study to see whether such a heightened response can protect against IAV infection needs to be evaluated both in vitro as well as in vivo challenge models.
