Abstract

Breast cancer is among the most common cancers, and accounts for a significant fraction of cancer burden in females. Commercially available treatments for breast cancer usually have side effects of varying degrees of severity associated with them, and also tend to be quite expensive. Plant based treatments can thus provide a cheaper alternative with less side effects. MCF-7 is a widely used human breast cancer cell line, which is ER positive, PR positive and HER2 negative. This cell line was chosen as the subject of our studies. Lantana camara is a flowering plant which is widespread in India, and has been found to have various medicinal properties, including anticancer effects. Thus, an extract was prepared from L. camara leaves, and its effects on MCF-7 cells were tested. In this study, the ethanolic leaf extract of L. camara was found to drastically affect the morphology of MCF-7 cells. The extract also showed a dose-dependent cytotoxic effect upon the cells. The leaf extract was also shown to cause nuclear fragmentation and DNA condensation in the cells. Treatment with the extract also caused changes in the cell cycle phase distribution of the MCF-7 cells, with an increase in subG1 and S phase population and a decrease in the G2/M population. The study also confirmed that L. camara extract causes cell death via apoptosis. Finally, the extract was separated into different fractions through HPLC, and the cytotoxic effect of one fraction of the extract upon MCF-7 cells was confirmed. The results of this study suggest that L. camara contains bioactive compounds with potent anticancer effects, which hold potential for future therapeutic applications.
