Abstract

Mesenchymal stem cells (MSCs) have rapidly gained attention for the treatment of numerous inflammatory and autoimmune diseases due to their distinct immunomodulatory and regenerative capacity. They can be used for autologous and allogenic transplantation due to their hypoimmunogenic nature, preventing them from triggering an immune reaction when transplanted. MSCs can be isolated from different sources, and based on the source, significant functional differences have been observed between various types of MSCs. Bone marrow is the most wellstudied and best-characterized source of MSCs, but it has several disadvantages. An appealing alternative choice of MSC sources are the neonatal tissues which are discarded as medical waste after birth and can be easily procured with informed consent from the donor. Wharton's jelly derived MSCs (WJ-MSCs) are of fetal origin and their isolation procedure has been wellestablished in our lab. In this study, we have established the isolation procedure for MSCs from human placenta (PLMSCs). For that, we chose the decidua basalis layer, which is derived from the maternal side of the placenta. We optimized an explant culture protocol for the isolation of the PL-MSCs followed by their extensive characterization. Furthermore, a comparative study was done between PL-MSCs and WJ-MSCs based on certain basic cellular parameters like cell area, population doubling time, cell adhesion, and cell migration.
