Abstract

Developing effective non-immunogenic and affordable wound care for treating simultaneously bleeding disorder and non-healing diabetic wound is indeed a challenging endeavour. Total wound care designing needs the integration of both haemostatic and therapeutic angiogenic property. Accident victims and wounded soldiers suffer from traumatic coagulopathy due to reduced blood clotting components. Diabetic wounds are ischemic in nature and face huge complications due to reduced blood flow, poor angiogenic progression caused by a protracted inflammatory phase and a high proteolytic environment. In order to address the unresolved medical problems of ischemic wound and traumatic coagulopathy, this thesis will address the development of safe wound care, inspired by nature. Here, we have designed fibrin-inspired and fibrin-mussel footpad protein inspired adhesive sealant and examined their physicochemical properties, biocompatibility and sealant-forming efficacy, both in vitro and in vivo. To handle the ischemic pathologies of non-healing diabetic wound, we have developed a library of growth factor based combination therapy. We combined angiogenic proteins and also both angiogenic and arteriogenic proteins and also synthesize their peptide-mimetics and evaluated their angiogenic potential and blood-vessel forming potential both in vitro and in vivo. Our designed peptides mimicking PDGF-BB and VEGF-A exhibited the best angiogenic property.
