Abstract

During the hypothermic storage of Red blood cells (RBCs) in a blood center, RBCs undergo many physical and biochemical changes known as storage lesions, which reduce the shelf life of RBCs; moreover, it can compromise transfusion efficacy. This study aims to evaluate the extent of oxidative stress in RBCs during their storage and evaluate key oxidative and membrane parameters during RBC storage, including glutathione redox status (GSH/GSSG), malondialdehyde (MDA) levels, hemoglobin oxygen saturation, and membrane fluidity. 20 mL of leukodepleted packed RBC units mixed with SAGM additive solution (n = 10) were stored under hypothermic conditions in a blood center for up to 42 days. Aliquots were aspetically collected at defined intervals to measure reduced (GSH) and oxidized (GSSG) glutathione levels, MDA concentration as a marker of lipid peroxidation, hemoglobin oxygen saturation, and membrane fluidity using DPH fluorescence anisotropy. Storage led to a progressive decline in GSH levels and an increase in MDA levels (p≤ 0.05), indicating increased oxidative stress and increased membrane lipid peroxidation. Hemoglobin oxygen saturation remained steady with storage duration. Fluorescence anisotropy of ghost RBCs exhibited a decrease with storage time, indicating an increase in membrane fluidity.
