Abstract

Lantana camara is a weed of the Verbenaceae family that has shown the presence of phytochemicals that exert anticancer effects against several cancer cell lines including MCF-7 and MDA-MB-231. The plant shows changes in phytochemical type and quantity based on its habitat and the seasonal condition it faces. The ethanolic leaf extract of Lantana camara, collected from the vicinity of the Indian Institute of Science Education and Research Kolkata, has demonstrated significant anticancer activity againstMCF-7 and MDAMB- 231 breast cancer cell lines. However, it has not been investigated for the different phytochemicals it holds. Based on this, we decided to investigate them specifically for bioactive compounds with anticancer potential. Through HPLC, we were able to isolate a fraction that showed high anticancer bioactivity against MCF-7,MDA-MB-231 and also showed cytotoxicity against HEK-293T cell lines. Its gene expression study in MCF-7 cell line showed that LCLE when used in crude formdid not alter p53 significantly with respect to the Vehicle control. The bioactive fraction that showed the highest cytotoxicity in the gene expression studies presented a reduction in the expression of p53, BCL-2 and BAX. When this fraction was studied using TLC and LC-MS/MS, we observed that the fraction consisted of two different compounds with the LC-MS/MS also revealing the presence of impurity. LC-MS/MS analysis conclusively showed that the masses of the two major compounds were 470.3 Da and 568.3 Da. Through 1-D Proton NMR, we were able to identify the possible compounds which have been reported in literature. Lantanolic acid (Molecular weight 470.3 Da) and Camaric acid (Molecular mass 568.3 Da) along with its functional isomer Lantanalic acid were identified to be the potential candidates of the bioactive fraction. 1-D Carbon NMR was done for further conclusive identification but yielded a poor Signal to Noise ratio due to which we were unable to give a confirmed result for the presence of the mentioned compounds. In summary, we identified a specific fraction in LCLE using HPLC that exhibited potent cytotoxic effects against MCF-7, MDA-MB-231, and HEK-293T cell lines. We demonstrated that both LCLE and the bioactive fraction influenced gene expression profiles in MCF-7 cells. Additionally, we characterized the fraction’s chemical composition and proposed possible structures for its components based on existing literature.
