Abstract
Uncontrolled bleeding from traumatic wounds of accident victims and soldiers causes huge no. of deaths. Patients having bleeding disorders like haemophilia, von Willebrand disease or afibrinogenemia are difficult to handle in surgery due to coagulopathic conditions. To address such traumatic coagulopathy and impaired blood clotting, there is an urgent need to engineer nature-inspired nanoscale peptide-based material to seal the wound. Self-assembled peptide based functional biomaterials are promising tools for biomedical applications. We aim to engineer fibrin inspired peptide-based sealants to facilitate efficient blood clotting to handle bleeding disorders of accident victim, injured soldiers and haemophilia patients. Our designed sealants are stabilized by lysine-aspartate ionic interactions and also by Nε(γ-glutamyl) lysine isopeptide bond mediated covalent interactions which actually mimics natural fibrinogen γ chain. In another system we mimic mussel foot-pad protein adhesive strategy by incorporating tyrosine residue to improve nano-mechanical force. All these designed sealants show stronger mechanical properties compared to indigenous fibrin. It also demonstrates antibacterial activity, hemocompatibility, biocompatibility and biodegradability. In future such robust materials have great prospect to be used by the clinicians or surgeons in case of an emergency bleeding.
