Abstract
The cell membrane is a highly dynamic system and membrane tension physically regulates many cellular processes. To understand the underlying non-active physical mechanisms, studies on cells need to be complemented with studies on model membrane systems. Plasma membrane spheres (PMS) are model membrane systems that lack cortical actin network and have lipid composition close to that of cells. We use Reflection Interference Microscopy to map membrane fluctuations in PMS and compare it with cell data. We have observed reduced temporal fluctuations and reduced spatial heterogeneity in PMS compared to cells. We think the reduced spatial heterogeneity in PMS is due to the absence of internal cytoskeletal structures.
