    Abstract

    The research work presented here deals with the synthesis of three structurally related Ru(II)-pcymene complexes with formula [RuII(η⁶-p-cymene)LX]X where, X = Cl (1), Br (2) and I (3) and elucidate the hydrolysis and DNA binding ability. All of the complexes can bind moderately with human serum albumin with the order of 10² which indicates moderate binding affinity. In acidic pH 6.7 the rate of hydrolysis increases compared to pH 7.4 which may enhance the toxicity in cancer cells. The binding of the complexes (1-3) with 9-EtG were detected in ¹H NMR. UV-Vis studies suggest that the complexes are more prone to bind with Guanine base (GMP). These binding studies indicate some degree of selectivity and the DNA targeting nature of the complexes, although the corresponding deactivation in presence of cellular thiols were also found. The studies show that the thiol of reduced glutathione can bind with ruthenium centre which may cause deactivation.
