Abstract

The key objective of this thesis is to design improved peptide based total wound care, especially for diabetic wound, which involves sealing the external wound by peptide based hemostats and also accelerate the healing process via therapeutic angiogenesis by growth factor mimetic angiogenic peptides. Traumatic injury causes huge number of deaths of civilian and soldiers and any kind of injury and surgery become fatal for patients having inherited bleeding disorder. Additionally, the non-healing wounds, especially diabetic-wounds, cause severe complications, since the impacted tissue often heals imperfectly leading to scarring. To handle traumatic coagulopathy, impaired blood clotting and impaired wound-healing, there is a critical need to develop nature-inspired biocompatible total wound care. Chapter 1 of this MS-thesis, describes the wound healing problem, especially in case of diabetic wound and also lists of therapeutic molecules available in the market. Chapter 2 describes our computational approach to develop improved peptide based sealants. Chapter 3 describes the structural characterization of VEGF mimetic peptide and in vivo wound healing studies of VEGF mimetic peptides in combination with two of our designed sealants. Summary chapter has summarized the key findings of this MS-thesis.
