Abstract

A random polymer containing Norbornene as a backbone to exert vigorous antibacterial activity was proposed. The polymer consists of three monomers: Monomer 1 contains a Glycine unit attached to a Norbornene derivative. The Glycine unit is liked with a 2000mg molecular weight Polyethylene glycol (PEG). The PEG unit enhances water solubility, inhibits bacteria growth, and imparts a biocompatibility effect to the polymer. For Monomer 2, a Norbornene derivative with an attached phenol group was synthesised. To this was attached 1, 5 dibromo pentane. The Br end in it would later be functionalised with triethylamine to give N⁺ unit, which exerts a strong membrane disrupting effect on the bacterial cell wall. Monomer 3 was designed with the same unit in the backbone as monomer 2, except that the side chain contained a Coumarin derivative instead of 1,5 dibromo pentane. Coumarin is an excellent bacterial membrane targeting agent & its fluorescence helps to keep track of the drug in Vivo. The Coumarin derivative was synthesized, but Monomer 3 couldn’t be synthesized. The synthesized compounds were all characterized by ¹H and ¹³C NMR spectroscopy. A compound with polymer properties and three side chains, each with individual antibacterial features, promises a solid & potent antibacterial combination.
