Abstract
Alzheimer’s Disease (AD) is a very common neurodegenerative disorder, characterized by presence of amyloid-beta plaques, neurofibrillary tangles, and marked by its cognitive impairment. Glutamate and -aminobutyric acid (GABA) are major excitatory and inhibitory neurotransmitters, respectively, and play a major role in cognitive functions of the individual. The disruption of neurotransmitter cycling associated with these neurotransmitters results in many pathological as well as psychiatric disorders. In vivo 13C NMR spectroscopy studies in rodents and human brain indicate that neurotransmitter energetics is supported by the oxidative glucose metabolism. The present study evaluates the neurochemical profile in APP-PS1 mouse model of Alzheimer’s following [6,6’-D2]glucose infusion. The incorporation of 2H in neurometabolites was assessed indirectly using 1H NMR spectroscopy with better sensitivity. Moreover over, it provides the advantages of 2H detection without the need for an specialized hardware in NMR spectrometer.
