Abstract

Lots of anti-cancer drugs are available in the market, but still we are lacking in treatment of cancer completely. Because, most of the drugs are poorly water soluble and there is an issue of biocompatibility and biodegradability. Drugs available in the market is lacking in the reaching to the target cells, amount of drug release to the target cells. We are also facing problem due to limited ability to monitor therapeutic responses. Recent work polymerizes norbornene monomer using Ring Opening Metathesis Polymerization (ROMP) and used as drug delivery of Doxorubicin, an anti-cancer drug. But, this backbone was biocompatible not biodegradable. So, our work was motivated from the above mentioned work. We used lactones based monomer, because of their biodegradability. I had prepared first DOXI-N₃, then monomer δ - valerolactone having propargyl group, with its copolymerization with ε - caprolactone (PVL-PCL copolymer) and attachment of phosphate moiety (PVL-PCL-PHOS copolymer) for Fe₃O₄ nanoparticles attachment. Attachment of iron nanoparticles is fine – tuned work of my project. DOXI-N3 was attached with PVL-PCL-PHOS copolymer using azide -alkyne click chemistry. For solubility in water PEG-N₃ was prepared and attached with same click chemistry in fine – tuned work. For the above work, I used Ring Opening Polymerization (ROP), because of its broad advantage like, mild reaction conditions (in some cases) and reasonable reaction time. Absence of reaction by‐products and can proceed in a “living” fashion. One more thing which we required in drug delivery system is controlled molecular weight, it can easily controlled in ROP.
