Abstract
Post translational modifications(PTMs) are changes that occur in the proteins after their synthesis which leads to changes in their functionality as well as their expression. These changes might be at the amino acid level where single/multiple amino acids undergo some sort of a chemical modification like oxidation/reduction or it can be at the protein level where the entire protein moiety gets attached to another protein as part of a signaling cascade like ubiquitination. One such class of enzymes which modifies proteins is methionine sulphoxide reductase(MSR) which controls the oxidative state of methionine (Met). Other than plants, MSRs are also found in bacterial, yeast and mammalian cells. MSRs have a role in stress tolerance and ageing due to their anti-oxidant properties. Tomato is one of the most important crops in our country and also serves as a model to study different signalling pathways. MSRs, as previous studies suggest, play a crucial role in abiotic and stress responses in plants. But MSRs have not yet been properly characterised or worked upon with respect to tomato which is the basis of my study. In this thesis, I have tried to characterise the MSR-Bs found in tomato through both in-silico and molecular biology methods and study their expression levels under various kinds of stress followed by mapping out their activity levels.
