Abstract

WJ-MSCs are mesenchymal stem cells derived from WJ portion of the umbilical cord. These cells are known for their easy availability without ethical issues, immunosuppressive nature, and high proliferation potential. These properties make them a potential source for clinical as well as research purposes. We have optimised a cryoprotectant solution i.e. 10% DMSO+0.2M sucrose for cryopreservation of umbilical cord tissue pieces. WJ-MSCs derived from these cryopreserved tissue pieces were found to retain the basic characteristics of MSCs i.e. they were plastic adherent in nature, maintained similar morphology and surface marker antigens as that MSCs derived from fresh tissue. No difference was seen in the chondrogenic and adipogenic differentiated WJ-MSCs obtained from the cryopreserved culture vs. fresh culture (from early and late passage WJ-MSCs). We have seen the presence and localisation of NMIIA and NMIIB in WJ-MSCs. Scratch wound healing assay and time lapse video analysis revealed that blebbistatin treated cells took longer to migrate than control WJ-MSCs. Analysis of cell adhesion and extracellular molecules showed that 14 genes were upregulated and 4 genes were downregulated. This also supports our live cell imaging data. If we get an idea that how cytoskeleton plays role in differentiation this will help us in designing methods in which we can efficiently differentiate the MSCs and this will help us in treatment of many diseases related to cartilage.
