Abstract

‘One-pot’ process has drawn lots of attention to understand and engineer glucose tolerant β- Glucosidase which may enable economic cellulose hydrolysis and lower biofuel production costs.. In this process, both solubilization of lignocellulosic biomass that is the pretreatment process and the biocatalytic hydrolysis of cellulose occur in a single step. Importantly, all the constituents present in the reaction mixture interact with each other. This thesis describes interactions between water and ionic liquid, enzyme and ionic liquid and enzyme and glucose. • Chapter 1 describes the conversion process of lignocellulosic biomass to biofuel. Particularly, we elucidate the role of different enzymes in production of biofuel. Additionally, we describe the role of various solvent on the stability and activity of the enzymes. Not only the enzyme, but also properties of ILs are modulated in presence of water. • Chapter 2 describes water mediated weakening of interionic interaction in binary mixture of water and 1-butyl-3-methylimidazolium tetrafluoroborate ([BMIM][BF₄]) IL by using both the quantum chemical calculation and molecular dynamics (MD) simulation. • In chapter 3, we propose a correlation between the interaction energy of the constituent of ILs and viscosity of the system. • Chapter 4 describes the molecular level understanding of structural alteration of B8CYA8 enzyme in various glucose concentration using molecular dynamic (MD) simulation. • Chapter 5 described the effect of concentration of substrate [p-Nitrophenyl--D-glucoside (p- NPGlc.)] on the specific activity of B8CYA8 enzyme. • Chapter 6 describes the inactivation of B8CYA8 activity in presence of 1-ethyl-3- methylimidazolium chloride ([EMIM][Cl]) IL. • Chapter 7 describes the future research plan which can be done from the outcome of the this.
