Abstract
This thesis is divided in to five chapters. First two are mainly introduction part. Third chapter describes about “Synthesis and assembly of peptides containing N-phenylglycine” which represents the synthesis of the hybrid peptides containing N-phenylglycine for noncanonical folding and formation of diverse supramolecular structures in the solid state. In crystals, the NPG-Phe-OMe self-assemble to form a supramolecular sheet like structure, and the Boc-NPG-Tyr-Aib-OMe exhibits a hairpin-like structure. Moreover, NPG-Phe-OMe shows flower like morphology, Boc-NPG-Tyr-Aib-OMe exhibits microspheres like morphology. Chapter 4 which is entitled “Aib modified Amyloid beta peptide fragment 24-26” represents the synthesis of the tripeptide containing valine, glycine, serine and α-aminoisobutyric acid by solution phase methods, their characterization by NMR, FT-IR and Mass spectrometry. The peptide fragment Aβ 24-26 self-aggregate into amyloid-like fibril. The Congo red binding assay shows that the peptide Boc-Val-Gly-Ser-OMe fibrils are readily stained with the dye and exhibits green gold birefringence under polarized light. The FT-IR studies reveal that the reported peptide fragment of Aβ 24-26 self-associate through intermolecular hydrogen bonds to form supramolecular antiparallel β -sheet like structure. However, the Aib modification inhibits the amyloid like fibril formation. Chapter 5 contains summary and perspective of this thesis.
