Abstract

This thesis deals with the development of simple fluorescent probes for the detection of various biologically and environmentally relevant analytes including hydrazine, sulfide ions and trivalent lanthanides. In addition, development of a straightforward synthetic strategy for the preparation of a series of 1,2,3-triazole-fused tetrahydropyrazine derivatives is also an aim of this project. Chapter 1 deals with the general introduction on the current research based on recent literature. Chapter 2 comprises of the synthesis of a pair of readily synthesizable pyrene- and anthracene-based turn-on emissive hydrazine-detecting chemodosimeters which are shown to selectively detect hydrazine in environmental water samples, live cells and live fruit-fly larvae. The detection mechanism suggests the formation of a five-membered heterocyclic ring upon attack of the hydrazine. Chapter 3 deals with the development and studies of a highly selective easy-to-prepare sulfide-detecting probe. This chemodosimeter displays a novel analyte detection strategy via the transformation of the profluorophore into fluorescent species in the presence of the analyte, sulfide. In Chapter 4, we demonstrate that curcumin, a naturally occurring, non-toxic, off-the-shelf, commercially available compound containing a single fluorophore, is capable of discriminating among fourteen trivalent lanthanide ions at pH 6.8 and 8.2 through multivariate analysis by measuring the emission and the absorption spectra of the probe after addition of lanthanides. For the non-specialists, an easy-to-handle tool has been devised, using a smartphone app. Chapter 5 justifies that simple ‘click’ chemistry also has its place in this thesis for its novel approach for the preparation of pharmaceutically relevant compounds, apart from its welldocumented role in chemosensory appliactions. In this work, pharmacophoric tetrahydro- [1,2,3]triazolopyrazine heterocyclic frameworks have been shown with their two-step synthetic cyclisation process from aromatic primary amines. The scope of this synthetic methodology was also explored with various amino acid-based synthons.
