Abstract
T cells play the central role of adaptive immune system by recognizing broad spectrum of antigen and harmful substances. The T cell receptor complex recognize the antigen and transduce the signal to interior of the cells resulting multiple effector function. This TCR receptor has six CD3 molecules which has 10 Immunoreceptor Tyrosine based activation motifs. Phosphorylation of these motifs is the first event after T cell activation initiation. But the role of this ITAM multiplicity still has not been understood till now. There is various model which tries to explain the mechanism of this ITAM multiplicity, but none have been successful describing all the phenomenon observed. In this study, the primary goal of the investigation was to understand the ITAM multiplicity in CD3 ζ molecule, which has three ITAM compared to other CD3 molecules with one ITAM. The pattern of phosphorylation was observed by phosphorylating the CD3 ζ with three tyrosine kinases, Abl kinase, Src kinase and Hck kinase. This result can be extrapolated to the CD3 ITAM multiplicity.
